Opsynvi (Macitentan/Tadalafil) is a prescription medication used to treat pulmonary hypertension. Pulmonary hypertension is a condition where the pulmonary arteries on the right side of the heart become thick and stiff, narrowing the space for the blood to flow. This causes increased blood pressure to develop in the lungs. The active ingredients in Opsynvi are Macitentan and Tadalafil. Macitentan belongs to a class of drugs called endothelin receptor antagonists. Tadalafil belongs to a class of medications called phosphodiesterase 5 (PDE5) inhibitors. Opsynvi works by relaxing the blood vessels and increasing blood supply to the lungs.

We currently offer the brand Opsynvi from Canada (available soon) manufactured by Janssen in the strength of 10 mg/40 mg film-coated tablets. The generic Macitentan/Tadalafil is not available.
Get medical help immediately if you have any of these signs or an allergic reaction while using Opsynvi. The most common side effects of Opsynvi may include the following:
• Dizziness;
• Headache
Contact your doctor right away if you have any of the severe side effects, including:
• Anemia;
• Muscle pain;
• Water retention
These are not all of the side effects of Opsynvi. Always contact your doctor if you notice any unusual symptoms.
You should not use Opsynvi if you are allergic to Macitentan, Tadalafil, or any ingredient in Opsynvi. Before you start using this medication, tell your doctor about your medical history, especially if you:
• Are breastfeeding or planning to breastfeed;
• Are pregnant or planning to be pregnant;
• Have an intolerance to certain sugars;
• Have anemia;
• Have kidney disease;
• Have liver disease
Tell your doctor about all your medications, including prescription, OTC, herbal supplements, and vitamins. There may be drug interactions between Opsynvi and other medicines. Tell your doctor if you are pregnant or planning to become pregnant. This drug can harm an unborn baby. Use effective birth control to prevent pregnancy while using Opsynvi. Inform your doctor if you are breastfeeding or planning to breastfeed. Macitentan passes into breast milk and it may affect a nursing infant. Do not breastfeed while using this medicine. Use this medicine exactly as your doctor prescribes. Opsynvi is a tablet. Swallow the tablets whole with water, and do not crush or chew. You can take this drug with or without food. Opsynvi can make you dizzy. Do not drive or do any activity that requires alertness until you know how the medicine affects you. Avoid grapefruit products when using this drug. Avoid alcohol with Opsynvi as it can increase the severity of the side effects. Store this drug at room temperature, away from light and moisture. Keep out of the reach of children.
